Vitamin B3 Cuts Glaucoma Risk by Two-Thirds in Largest Real-World Study

Patients with elevated eye pressure who took nicotinamide – an over-the-counter form of vitamin B3 – were two-thirds less likely to develop the most common form of glaucoma than matched patients who took none, according to the largest real-world study to date to examine that relationship.

Published in the JAMA Ophthalmology journal, this study found that people with raised eye pressure who took nicotinamide, a form of vitamin B3, were around two-thirds less likely to develop the most common form of glaucoma than those who did not take it. They were also less likely to need prescription eye drops or laser treatment. Since glaucoma is a leading cause of irreversible blindness and often develops without warning signs, the findings have attracted considerable interest.

Scientists believe the benefit may come from vitamin B3’s ability to support the health of the optic nerve. As people age, the body’s levels of a vital energy-producing molecule known as nicotinamide adenine dinucleotide (NAD+) naturally decline, making nerve cells in the eye more vulnerable to damage caused by high eye pressure. Nicotinamide helps restore NAD+ levels, allowing these cells to function more efficiently and better resist damage. Earlier laboratory studies and small clinical trials have already suggested that vitamin B3 could help protect vision, and these latest findings add further support to that idea.

Several major clinical trials are now underway, with results expected by the end of 2026. If these studies confirm the current findings, vitamin B3 could become an important new approach for helping to prevent one of the world’s leading causes of blindness.
